ABSTRACT

PURPOSE: Recent evidence suggests that B cells can both promote and inhibit the development and progression of allergic disease. However, the characteristics of B cell subsets in patients with allergic rhinitis (AR) have not been well documented. This study aimed to analyze the characteristics of B cell subsets in the peripheral blood of AR patients. METHODS: Forty-seven AR patients and 54 healthy controls were enrolled in this study, and the B cell subsets in peripheral blood of all subjects were analyzed by flow cytometry. Moreover, the serum total immunoglobulin E (IgE) and IgE concentrations secreted into the cultured peripheral blood mononuclear cells (PBMCs) were measured by using enzyme-linked immunosorbent assay. RESULTS: We found the peripheral blood of AR patients contained higher percentages of memory B cells, plasma cells, and CD19+CD24hiCD27+ regulatory B cells (Bregs) than those of age-matched healthy controls (P < 0.05), while the percentages of naïve B cells and CD19+CD24hiCD38hi Bregs were significantly lower in AR patients than in healthy individuals (P < 0.05). In addition, the serum total IgE and IgE concentrations secreted into the cultured PBMCs were elevated in AR patients than in the healthy controls (P < 0.05). CONCLUSIONS: Our findings indicate that AR patients were characterized by increase in terminally differentiated memory B cells or plasma cells and decreases in CD19+CD24hiCD38hi Breg cells in the peripheral blood.

ABSTRACT

A new type of embedded acupoint recognition device was developed for acupoint recognition and teaching. Through improving the acupoint electric resistance detection technique of the two-electrode method, combined with mechanical structure design and embedded control technology, the human acupoint database was assembled in the embedded chip with the international symbol, acupoint name, , location, chart, manuscript and audio information included, a new type of embedded acupoint recognition device was designed. The device was composed of 3 modules, named the acupoint electric resistance detection system, the embedded control system and mechanical system. It achieved skin contact sensation, the collection of electric resistance at the required skin region, the wireless communication, the liquid crystal display (LCD) human-computer interaction and acupoint magnetic head massage. When used, the resistance value of the detected skin is read on LCD to determine whether the detected skin resistance meets the features of low resistance of acupoint and further to identify the acupoint zone. Afterward, the data collected are matched with the human acupoint database to determine the information of acupoint. The control mechanical structure achieves the acupoint pressing and kneading. After verified by many operation staff, it is shown that the device not only improves the recognition speed and precision of acupoint, but also contributes to the acupoint teaching with the human acupoint database displayed on LCD. The achievements benefit the promotion of meridians and acupoints as well as the development of acupuncture devices.

ABSTRACT

T cell immunoglobulin and mucin domain 3 (Tim-3) is well known to negatively regulate T cells responses, but its role in burn-induced T cells immune suppression remains unclear. In the present study, in order to identify the relationship between Tim-3 expression and post-burn T cells immune suppression, C57BL/6 mice were subjected to burn injury or sham injury, and the liver and spleen were harvested at the day 1 after operation. The expression level of Tim-3 on hepatic or splenic T cells and the functional properties of Tim-3(+) T cells were evaluated. It was found burn injury induced dramatically elevated Tim-3 expression on both hepatic and splenic CD4(+) and CD8(+) T cells in contrast with the post-burn depletion of T cells. Furthermore, Tim-3 expression was correlated with the suppressive phenotype of T cells following burn injury, including increased expression of anti-inflammatory cytokine IL-10, decreased expression of pro-inflammatory cytokines IFN-γ and TNF-α, reduced T cell proliferation and elevated co-expression of Tim-3 and PD-1. Moreover, Tim-3(+) T cells subsets were more prone to spontaneous apoptosis than Tim-3(-) T cells subsets. Our findings reinforce the idea that the up-regulated expression of Tim-3 on T cells after burn injury plays an important role in the development and maintenance of burn-induced T cell immune suppression.

ABSTRACT

<p><b>OBJECTIVE</b>To clarify the relationship between the loss of expression of the deleted in pancreatic carcinoma locus 4 (DPC4) proteins and the pathogenesis of biliary tract carcinoma.</p><p><b>METHODS</b>71 primary biliary tract carcinoma (BTCa), including 38 common bile duct (CBD) carcinomas, 18 gallbladder carcinomas, 15 hilar bile ducts (HBD) carcinomas were examined by immunohistochemical staining. In addition, the CBD carcinomas were divided into two groups: tumors with metastasis (M(+) group, 27 cases) and tumors without metastasis (M(-) group, 11 cases).</p><p><b>RESULTS</b>The frequency of loss of the expression of DPC4 protein was 32.8% in BTCa, 47.3% in CBD carcinoma, 11% in gallbladder carcinoma, 13% in HBD carcinoma. Comparison of the frequency of loss expression of DPC4 was significant statistical difference in CBD carcinoma versus gallbladder carcinoma and HBD carcinoma (P < 0.01). The frequency of loss expression of DPC4 was 48.1% in the M(+) group and 45.4% in the M(-) group.</p><p><b>CONCLUSION</b>There are a close relationship between pathogenesis of BTCa and inactivation of DPC4 and different frequencies of DPC4 gene alternation in various locations of the biliary tract, which are not significantly increased with tumor metastasis in BTCa.</p>

ABSTRACT

Objective To observe the effect of a specific T-cell induced by lysate-pulsed human dendrtic cells against pancreatic cancer(PC) cells . Methods Dendritic cells were amplified and purified from peripheral blood of PC patient and were pulsed with tumor cells.Then they were co-cultured with autologus T-cell in vitro and divided into lysate-pulsed DCs group, unplused DCs group, lysates group,and control group. The supernatants of every co-culture group were collected and the concentration of IL-12 and IFN-? were measured by ELISA. The capacity of DC induced proliferation of T-cells was tested.The specific cytolytic activity of CTL was assessed in a 4-hr 51 Cr-release assay. Results Difference in the concentration of IL-12 and IFN-? was statistically significant in lysate-pulsed DCs group(1161?239 pg/ml and 1044?312 pg/ml)than these in unplused DCs group and lysates group (P

The controlling system of triple pulse vacuum sterilizer for dental use adopts PIC single-chip computer as the controller. The information related to stream temperature and pressure of the stream and liquid mixture in the hermetic vessel is acquired and controlled. Linearization measurements of the temperature and pressure and fuzzy control technology are all involved in. The experiment result shows that the controlling system makes the sterilizer free from overshoot, steady-state error and non-robustness.

ABSTRACT

Objective To understand the RHD gene profiles of RhD negative individuals in Hainan Han population,and provide reference to establish the right method for RHD genotyping.Methods RhD was tested by anti-globulin test, and RhDel and genuine RhD-negative phenotype were identified by absorption/elution method. RhD negative samples were further tested for RHD exons by PCR-SSP.The RhD negative samples with intact RHD genes were further analyzed by PCR-SSP for RHD introns 2, 10 and RHD?gene. Results Thirty-one (29.25%)cases of RhDel individuals were identified among 106 apparent RhD-negative individuals. All RhDel samples had RHD genes;67 cases of genuine RhD-negative had no RHD genes and 8 cases were partial D. All 31 RhDel samples had Din2 and Din10 but none had RHD?. Additionally, We detected exons 1,3,4,6,7,9 and 10 in one case of ccdEe sample. Conclusion The proportion of RhDel phenotype is high among apparent RhD-negative Hainanese, and total RHD exons can be detected in all RhDel samples. Polymorphisms of RHD gene are present among genuine RhD-negative Hainanese. There is no exon 5 in all 8 cases of partial D, which suggests that exon 5 specific amplification may be very important in RHD genotyping for Hainan Han population.
